检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人MMP-3抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的MMP-3与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中MMP-3的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长5632 nm)测定吸光度值。

MMP3 分子靶点综述
基质金属蛋白酶3(MMP-3),又名基质分解素-1,在人类中由MMP3基因编码。MMP3的表达主要在转录水平上调控,转录水平上的基因启动子对多种刺激物包括生长因子、细胞因子、促癌剂和原癌基因产物作出应答。MMP-3酶降解II、III、IV、IX和X型胶原蛋白、蛋白聚糖、纤维连接蛋白、层粘连蛋白和弹性蛋白。另外,MMP-3还可激活其它MMPs如MMP-1、MMP-7和MMP-9,因此MMP-3在结缔组织重塑中起关键作用。该酶参与伤口修复、动脉粥样硬化进展和肿瘤启动。MMP-3野生型和基因敲除的小鼠研究表明,MMP-3增加了外伤性损伤后的血脑屏障通透性。细胞核MMP-3可作为转录因子和蛋白酶发挥作用。

Involved in several processes, including cellular response to UV-A; negative regulation of hydrogen peroxide metabolic process; and positive regulation of protein-containing complex assembly.
