检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠TSLP抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的TSLP与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中TSLP的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

TSLP 分子靶点综述
胸腺基质淋巴细胞生成素(TSLP)主要由非造血细胞如成纤维细胞、上皮细胞和不同类型的基质或基质样细胞产生。它主要影响骨髓细胞、诱导单核细胞释放T细胞虏获趋化因子和增强髓样(CD11c+)树突状细胞成熟。研究表明,TSLP还可激活位于表皮的特定树突状细胞亚群—朗格汉斯细胞的成熟。胸腺TSLP激活髓样和浆细胞样(CD123+)树突状细胞会导致调节性T细胞的产生。TSLP通过胸腺基质淋巴细胞受体CRLF2和IL-7Rα链组成的异源二聚体受体复合物产生信号。结合STAT5后就会诱导其发生磷酸化,进而导致上游转录因子的表达。TSLP的表达与许多疾病包括哮喘、炎症性关节炎、特应性皮炎、湿疹及其它过敏状态有关。
