检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠/大鼠IGF-1抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品和待测样本,经过孵育,样本中存在的IGF-1与固相抗体结合。洗涤去除未结合的物质后,加入生物素化的检测抗体孵育。洗涤去除未结合的生物素化的抗体,加入辣根过氧化物酶标记的链霉亲和素 (Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中IGF-1的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

IGF1 分子靶点综述
胰岛素样生长因子1(IGF-1),也称为生长调节素C,是一个在分子结构上类似于胰岛素的激素。IGF-1主要由肝脏产生,作为内分泌激素。同样地,靶组织亦能旁分泌/自分泌产生IGF-1。IGF-1是AKT信号转导途径中的一种有效天然激活剂、细胞生长和增殖的刺激子、细胞程序性死亡的有效抑制子。IGF-1最少可结合两种细胞表面受体:IGF-1受体(IGF1R)和胰岛素受体。IGF-1的产生受生长激素刺激,受营养不良、生长激素不敏感、缺少生长激素受体或者生长激素受体(SHP2和STAT5B)下游信号通路发生故障等方面所抑制。IGF-1在儿童生长过程中起重要作用,继而在成人合成代谢中有影响,且与老年化、神经病变、癌症以及中风相关。

Involved in negative regulation of gene expression; negative regulation of release of cytochrome c from mitochondria; and positive regulation of myelination. Acts upstream of or within several processes, including animal organ development; positive regulation of cell population proliferation; and regulation of signal transduction. Located in exocytic vesicle and extracellular space. Is expressed in several structures, including alimentary system; central nervous system; genitourinary system; limb; and sensory organ. Human ortholog(s) of this gene implicated in several diseases, including acne; artery disease (multiple); diabetes mellitus (multiple); myopia; and neurodegenerative disease (multiple). Orthologous to human IGF1 (insulin like growth factor 1).
