检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠IL-17F抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的IL-17F与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中IL-17F的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

IL17F 分子靶点综述
白细胞介素17F(IL-17F)和IL-17A是IL-17家族中紧密相关的两个成员,有大约50%氨基酸同源。IL-17F来源于不同细胞类型,如Th17细胞、肥大细胞和嗜碱性粒细胞,在组织中广泛表达,包括肺。IL-17F可诱导支气管上皮细胞、血管内皮细胞、成纤维细胞和嗜酸性粒细胞中的许多细胞因子、趋化因子和粘附分子。IL-17F利用IL-17RA和IL-17RC作为其受体,激活MAP激酶相关通路。小鼠气道IL-17F基因的过表达与气道中的中性粒细胞增多、多种细胞因子的诱导、气道高反应性和粘液高分泌增加有关。因此,IL-17F在过敏性气道炎症中可能起到重要作用,对哮喘有重要的治疗意义。
