检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人TIMP-1抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的TIMP-1与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中TIMP-1的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

基质金属蛋白酶抑制剂-1(TIMP-1)是一种在生物体多种组织中表达的糖蛋白,是TIMP家族中的一员。TIMP-1是一种天然的基质金属蛋白酶(MMPs)抑制剂,后者是参与细胞外基质降解的一组肽酶。TIMP-1除了可抑制大多数已知的MMPs外,还可促进多种细胞类型的细胞繁殖,也可能具有抗凋亡作用。对许多细胞因子和激素的应答可强烈诱导该基因的转录。研究表明,人类女性中的TIMP-1基因失活是多态性的。肾上腺皮质细胞的营养激素ACTH诱导TIMP-1的表达,且TIMP表达的增加与胶原酶活性的降低有关。另外,TIMP-1表达的增加与多种肿瘤如喉癌或黑色素瘤的预后较差有关。
