检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人Serpin E1抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的Serpin E1与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中Serpin E1的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

Serpin E1,又名纤溶酶原激活物抑制因子-1 (PAI-1),是丝氨酸蛋白酶抑制剂超家族中的一员,主要由内皮(血管内皮衬里细胞)产生,但也由其他类型的组织如脂肪组织分泌产生。Serpin E1是尿激酶和组织型纤溶酶原激活剂(uPA和tPA)的主要抑制剂。PA-血纤维蛋白溶酶系统参与多种生理和病理过程如纤维蛋白溶解、纤维化、血管生成、伤口愈合、肿瘤细胞的侵袭和转移。例如,高水平的uPA和Serpin E1与乳腺癌患者的预后不良相关,研究证实uPA和Serpin E1的临床价值处于1级水平,是最可靠的生物标志物。除了乳腺癌,Serpin E1也参与许多其它疾病如代谢综合征、哮喘、重度抑郁症(MDD)以及衰老相关疾病。血浆中Serpin E1的缺失将导致异常出血,高水平的Serpin E1与遗传性易栓症相关。
