检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人IL-23抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的IL-23与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中IL-23的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

IL23A 分子靶点综述
白细胞介素23(IL-23)是一种由IL-12p40亚基和IL-23p19亚基组成的异二聚体细胞因子,其中p40亚基是IL-23与IL-12共用的。IL-23的功能性受体已被鉴定,由IL-12Rβ1和IL-23R组成。IL-23在1型极化T细胞的免疫应答中起作用。虽然IL-12可强有力激活幼稚T细胞,但是最初的报道认为IL-23可优先作用于记忆T细胞,促进其分泌IFN-γ和增殖,这表明IL-23在控制细菌感染上有重要作用。IL-23被进一步描述为是一种控制周围组织炎症的关键细胞因子。p19的过表达与多个器官以及包含皮肤在内的上皮组织的炎症有关。IL-23参与中枢神经系统和自身免疫性疾病的炎症。
