检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人IL-12p70抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的IL-12p70与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中IL-12p70的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

IL12B 分子靶点综述

白细胞介素12 (IL-12)由树突状细胞、巨噬细胞、中性粒细胞和人B淋巴母细胞株 (NC-37)在响应抗原刺激时自然产生。IL-12由4个а螺旋组成,是一个由2种独立基因--- IL-12A (p35)和IL-12B (p40)编码形成的异源二聚体细胞因子。蛋白合成之后可形成有生物活性的异源二聚体(p70)和同源二聚体p40。IL-12参与初始T细胞向Th1细胞分化的过程,作为T细胞刺激因子,它可刺激T细胞的生长及其功能发挥。IL-12具有抗血管生成活性,对自然杀伤细胞和T淋巴细胞活性有重要作用。同样地,IL-12与自身免疫相关。
