检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人CCL4抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的CCL4与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中CCL4的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

CCL4 分子靶点综述
趋化因子配体4(CCL4),又名巨噬细胞炎性蛋白1β(MIP-1β),是一种对CCR5受体具有特异性的CC趋化因子。它能趋化自然杀伤细胞、单核细胞和其它多种免疫细胞,可被小鼠的许多细胞类型诱导表达,包括肥大细胞、内皮细胞、巨噬细胞和CD8+T细胞。小鼠CCL4的cDNAs编码一段含有23个氨基酸残基信号肽的92个氨基酸残基前体蛋白,裂解该前体蛋白可产生69个氨基酸残基的分泌性的成熟蛋白。CCL4是一种由CD8+T细胞产生的重要的HIV抑制因子。趋化因子的活性由G蛋白偶联的7次跨膜结构域受体调节,CCR1、5和8这些受体可调节CCL4的生物活性。在同一细胞群中,CCL4的活性比CCL3弱20倍。
