检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人BDNF抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的BDNF与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中BDNF的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

BDNF 分子靶点综述

脑源性神经营养因子(BDNF)是生长因子神经营养素家族中的一员,与神经生长因子相关。神经营养因子存在于脑及外周。BDNF在内质网中加工合成,由致密核心囊泡分泌。它在大脑中的海马、皮层和基底前脑区域活跃,同样也在视网膜、运动神经元、肾脏、唾液和前列腺中表达。BDNF作用于中枢神经系统和外周神经系统的某些神经元,帮助现存神经元的存活、促进新生神经元和突触的生长和分化。BDNF的改变可能在精神分裂症的发病中起作用,与抑郁、湿疹、癫痫、阿尔茨海默病、肥胖、衰老、先天性中枢性低通气综合征和化疗后认知功能障碍有关。
