使用方法

1.收集细胞，调整细胞浓度到10⁶ -10⁷个/ml。
2. 在流式管中加入 50 μl 细胞，加入表染抗体。涡旋震荡混匀，室温避光孵育 15 分钟。
3. 每管加入 2 ml 1× FCM Lysing Solution 工作液，涡旋震荡混匀，室温避光孵育 15 分钟。室温下 300 - 400 × g 离心 5 分钟，弃去上清。
4. 每管加入 2 ml 1× Flow Cytometry Staining Buffer，涡旋震荡混匀。室温下 300 - 400 × g 离心 5分钟，弃去上清。
5. 每管加入 1 ml 1× Fixation/Permeabilization 工作液，涡旋震荡混匀。
6. 室温避光孵育 30 - 60 分钟。
7. 无需洗涤，每管加入 2 ml 1× Permeabilization Buffer。室温下300 - 400 × g 离心 5 分钟，弃去上清。
8. 用100 μl 1× Permeabilization Buffer重悬沉淀。通常洗涤后残留的液体量就已足够。
9.加入5 μl Anti-Rat Foxp3, PE或5 μl Rat IgG2a Isotype Control, PE。震荡混匀，室温避光孵育至少30分钟。
10.每管加入2 ml 1× Permeabilization Buffer，室温下300 - 400 × g离心5分钟，弃去上清。
11.每管加入 500 μl 1× Flow Cytometry Staining Buffer 重悬，上机检测。
