ITGAM 分子靶点综述
整合素αM(ITGAM)是一种形成巨噬细胞抗原(Mac-1)或补体受体3(CR3)的蛋白亚单位。ITGAM又被称为CR3A和分化抗原簇分子11B(CD11b)。CD11b是一种常见的髓系标志物,在急性髓系白血病中有50%的阳性表达率。CD117和CD11b共同检测有助于从恢复良性增殖(CD117阴性表达,CD11b阳性表达)的病例中区分出急性早幼粒细胞白血病(CD11b阴性表达)。

Involved in several processes, including central nervous system development; endocytosis; and positive regulation of neuron death. Acts upstream of or within several processes, including activated T cell proliferation; leukocyte migration; and microglia development. Located in external side of plasma membrane and nucleus. Is integral component of membrane. Part of integrin alphaM-beta2 complex. Is expressed in several structures, including central nervous system; heart; lymphatic vessel; thymus; and yolk sac. Human ortholog(s) of this gene implicated in lupus nephritis. Orthologous to human ITGAM (integrin subunit alpha M).

使用方法
全血样本:取100 μl 抗凝血,加入5 μl Anti-Human/Mouse CD11b, FITC 和其他抗体,震荡混匀,于室温避光孵育15 分钟或2 - 8℃避光孵育30 分钟。根据红细胞裂解液说明书进行操作,去除红细胞。300×g 离心 5 - 10 分钟,弃上清,加入 500 μl 流式染色缓冲液重悬,上机检测。
其他样本:将 1×105 - 108 个单细胞经适当离心后,取适量流式染色缓冲液 (目录号S1001)重悬,加入5 μl Anti-Human CD11b, FITC 和其他抗体,使反应终体积为 100 μl。震荡混匀,于室温避光孵育15 分钟或 2 - 8℃避光孵育 30 分钟。每管加入 1 - 2 ml 流式染色缓冲液,300 × g 离心 5 - 10 分钟,弃上清,加入 500 μl 流式染色缓冲液重悬,上机检测。
实验中,可对抗体进行优化滴定以获得最佳效果。
