CD274 分子靶点综述

程序性死亡配体1(PD-L1)，也称为分化簇274(CD274)或 B7同源物1(B7-H1)，是由CD274基因编码的一种分子量为40kDa的I型跨膜蛋白。PD-L1在妊娠、组织移植、自体免疫性疾病以及其他疾病如肝炎的免疫系统抑制过程中可能发挥着重要作用。通常情况下，免疫系统对外来抗原产生反应，这些抗原积聚在淋巴结或脾脏中，引发抗原特异性 CD8+ T 细胞的增殖。PD-1受体/PD-L1或B7.1受体/PD-L1结合传递抑制信号，从而抑制 CD8+ T 细胞的增殖。相反，有报道表明PD-L1对休眠T细胞具有共刺激作用。PD-L1可能是通过诱导T细胞的凋亡和阻断细胞周期进程，从而抑制T细胞的持续反应。
