APOE 分子靶点综述

载脂蛋白E (Apo E)是一类在乳糜微粒和中间密度脂蛋白中发现的载脂蛋白，对于富含甘油三酯的脂蛋白成分的正常代谢是必不可少的。外周组织中，Apo E主要由肝脏和巨噬细胞产生，介导异构体依赖性的胆固醇代谢。中枢神经系统中，Apo E主要由星形胶质细胞产生，可通过Apo E受体运输胆固醇到神经元。Apo E运输脂蛋白、脂溶性维生素和胆固醇进入淋巴系统，然后进入血液。在免疫调节领域，越来越多的研究指出Apo E与许多免疫过程(包括抑制T细胞增殖、调节巨噬细胞功能、促进脂质抗原递呈给自然杀伤T细胞、调节炎症和氧化)相互作用。由单核细胞分泌的Apo E受炎性细胞因子下调，受TGF-β上调。该蛋白参与阿尔茨海默病和心血管疾病。
