检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠MMP-9抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、检测样本和辣根过氧化物酶标记的检测抗体,经过孵育,样本中存在的MMP-9与固相抗体和检测抗体结合。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中MMP-9的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

MMP9 分子靶点综述
基质金属蛋白酶9(MMP-9),又名92kDaIV型胶原酶、92kDa明胶酶或者明胶酶B,是一类属于锌-金属蛋白酶家族的酶。MMP-9参与正常生理过程(如胚胎发育、繁殖、血管形成、骨骼发育、伤口愈细胞迁移、学习和记忆)和病理过程(如关节炎、脑出血和肿瘤转移)的细胞外基质降解。MMP-9和弹性蛋白酶似乎是中性粒细胞迁移通过基底膜的调节因子。MMP-9在血管和新生血管形成中起重要作用。它在人类呼吸道上皮愈合过程中被显著上调,并被发现与许多病理过程相关,如癌症、免疫和心血管疾病。在类风湿性关节炎、局灶性脑缺血案例中可发现MMP-9的水平是升高的。它的水平随着特发性房颤的发展而增加。

Acts upstream of or within several processes, including collagen catabolic process; embryo implantation; and extracellular matrix organization. Located in collagen-containing extracellular matrix. Is expressed in several structures, including alimentary system; central nervous system; early conceptus; limb mesenchyme; and skeleton. Human ortholog(s) of this gene implicated in several diseases, including Down syndrome; anodontia; artery disease (multiple); autoimmune disease (multiple); and lung disease (multiple). Orthologous to human MMP9 (matrix metallopeptidase 9).
