产品简介
本产品用于检测小鼠抗凝血或脾细胞中,Th17细胞在T淋巴细胞 (或CD4+辅助T细胞)中的比例。通常所说的Th1、Th2和Th17是指在各种生理与病理条件下有能力分化为Th1、Th2和Th17 的T辅助细胞 (严格意义上是Th0细胞)。静息状态 (即未受任何刺激,如小鼠的正常生理状态)下,Th0分化为Th1、Th2和Th17的能力非常弱,所以外周血中仅含有极少量的Th1、Th2和Th17细胞,这时所能检测到的IFN-γ、IL-4和IL-17A也微乎其微。而当Th细胞受到外界因素 (如刺激素、病原体等)刺激,其中Th0即会向Th1、Th2或Th17分化,具体分化趋向取决于细胞因子的种类。此时,检测到的IFN-γ、IL-4或IL-17A也较多。实验中检测的Th1、Th2和Th17实际上是检测Th细胞对刺激素刺激的反应能力。

Flow cytometric analysis of Mouse Th17 Staining Kit. The staining pattern of IL-17A on resting heparin anticoagulated blood (A0), PBMCs from EDTA anticoagulated blood (B0), splenocytes (C0) and spleen mononuclear cells (D0 ) of normal ICR mouse. PMA/Ionomycin stimulated heparin anticoagulated blood (A1), PBMCs from EDTA anticoagulated blood (B1), splenocytes (C1) and spleen mononuclear cells (D1) of normal ICR mouse. Dot plot analysis are derived from gated CD3+/CD4+ cells population. Flow cytometry was performed on a Thermo Fisher Attune NxT.
使用Mouse Th17 Staining Kit进行流式检测。正常ICR小鼠的静息肝素抗凝血(A0)、EDTA抗凝血来源的PBMCs (B0)、脾细胞(C0)和脾单个核细胞(D0)染色IL-17A。正常ICR小鼠的PMA/Ionomycin刺激的肝素抗凝血(A1)、EDTA抗凝血来源的PBMC (B1) 脾细胞(C1)和脾单个核细胞(D1)染色IL-17A。对CD3+/CD4+细胞进行设门分析。实验在Thermo Fisher公司的Attune NxT流式细胞仪上进行。
