线粒体电化学势梯度(ΔΨ)丧失是凋亡早期的事件。本试剂盒提供一种快速方便的方法,利用阳离子亲脂性染料JC-1 (5,5’,6,6’-tetrachloro-1,1’,3,3’- tetraethylbenzimidazolocarbocyanine iodide)检测活细胞线粒体膜电位的变化,适合于凋亡细胞的检测。在正常细胞中,由于电化学势梯度,JC-1积聚在线粒体基质中,形成红色荧光的聚集体。破坏线粒体膜电位的事件会阻止JC-1染料积聚在线粒体中,因此该染料会扩散到整个细胞,使红色荧光转变为绿色荧光(JC-1单体)。CCCP (carbonyl cyanide 3-chlorophenylhydrazone)可用于研究线粒体膜电位。JC-1在线粒体中的积聚依赖于电位,其发射光从绿色(~529 nm)迁移到红色(~590 nm)。因此,线粒体去极化时,红色/绿色荧光强度之比会下降。JC-1在多种类型的细胞包括肌细胞和神经元,以及完整的组织和分离的线粒体中都可以作为线粒体膜电位的指示剂。
