检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人sVCAM-1抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的sVCAM-1与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中sVCAM-1的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

VCAM1 分子靶点综述
血管细胞粘附分子1(VCAM-1)也称为CD106,是100-110kDaI型跨膜蛋白,通常具有7个C2型免疫球蛋白结构域,作为细胞黏附分子发挥功能。VCAM-1蛋白介导淋巴细胞、单核细胞、嗜酸性粒细胞和嗜碱性粒细胞等黏附于血管内皮。VCAM-1是整合素β1亚家族VLA-4(极迟抗原-4或α4β1)和整合素α4β7的内皮配体。在其他类型细胞上也观察到VCAM-1的表达。它也能与EZR和膜突蛋白作用。VCAM-1的其它功能包括通过细胞-细胞接触调控破骨细胞生成、血氧不足时诱导镰状细胞黏附于血管内皮细胞。它还在白细胞-内皮细胞信号传导中发挥作用。监测血清中的sVCAM-1表达水平可为多种病理状态提供更详细的视角,如动脉粥样硬化和类风湿性关节炎。

Involved in several processes, including calcium-mediated signaling using intracellular calcium source; cell-cell adhesion in response to extracellular stimulus; and cellular response to amyloid-beta. Located in several cellular components, including early endosome; external side of plasma membrane; and podosome. Part of alpha9-beta1 integrin-vascular cell adhesion molecule-1 complex. Biomarker of several diseases, including artery disease (multiple); diabetes mellitus (multiple); essential thrombocythemia; glomerulonephritis (multiple); and interstitial cystitis.
