检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人sE-Selectin抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的sE-Selectin与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中sE-Selectin的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

SELE 分子靶点综述
E-选择素,又名CD62E、内皮-白细胞粘附分子1(ELAM-1)或者白细胞-内皮细胞粘附分子2(LECAM2),是一个115kDa的糖蛋白,只有被炎性细胞因子(IL-1β或TNF-α)或内皮细胞中的内毒素激活后才表达。细胞表面的E-选择素介导白细胞附着到内皮,从而在局部炎症反应中起重要作用。E-选择素的胞外区包括一个钙依赖性的C2型凝集素结构域、一个表皮生长因子(EGF)结构域和6个重复的补体调节蛋白样序列。可溶性E-选择素(sE-Selectin)存在于健康机体的血液中,可能产生于表面表达分子的蛋白裂解。监测血清中sE-Selectin水平可为一些病理情况如癌症、感染性疾病、糖尿病和脓毒病提供更详细的见解。
