检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人OPN抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品和待测样本,经过孵育,样本中存在的OPN与固相抗体结合。洗涤去除未结合的物质后,加入生物素化的检测抗体孵育。洗涤去除未结合的生物素化的抗体,加入辣根过氧化物酶标记的链霉亲和素 (Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中OPN的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

SPP1 分子靶点综述
骨桥蛋白(OPN),又名BSP-1、ETA-1、SPP-1或Ric,在人类中由SPP1基因编码,主要在骨骼中表达。OPN基因的调节机制还未完全阐明,不同细胞类型的调节机理是不同的。OPN在生物矿化、骨重塑、免疫功能、趋化、细胞活化和凋亡上有作用,同样与心肌细胞凋亡和心肌功能紊乱的增加有关。OPN与许多生理和病理过程包括伤口愈合、骨转换、肿瘤发生、炎症、缺血和免疫应答有关。调控血浆(或局部)的OPN水平对治疗自身免疫性疾病、癌症转移、骨和牙齿的矿化、骨质疏松症和一些应激形式有帮助。

Located in Golgi apparatus and extracellular space.
