检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人LIF抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品和待测样本,经过孵育,样本中存在的LIF与固相抗体结合。洗涤去除未结合的物质后,加入生物素化的检测抗体孵育。洗涤去除未结合的生物素化的抗体,加入辣根过氧化物酶标记的链霉亲和素 (Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中LIF的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

LIF 分子靶点综述

白血病抑制因子(LIF)是一种白细胞介素6家族细胞因子,通过抑制分化影响细胞生长。当LIF水平下降时,细胞就会分化。活化的T细胞、单核细胞、星形胶质细胞、成骨细胞、角质形成细胞、肥大细胞和成纤维细胞都可表达LIF。LIF因其能诱导髓系白血病细胞的终端分化,阻止白血病细胞的持续增长而得名。LIF的其它特性包括促进生长、促进不同类型的靶细胞分化、影响骨代谢、恶病质、神经发育、胚胎发育和炎症。LIF参与孕期子宫内膜蜕膜化和胚胎植入子宫内膜的过程。女性体内的LIF和其它细胞因子水平下降时,就容易怀孕,但是不明原因复发性流产的风险会增加。研究表明重组人LIF可能有助于改善女性不明原因不孕不育的着床率。另外,LIF通常用来添加到干细胞培养基中来减少自发分化。
