检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人IL-15抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的IL-15与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中IL-15的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

白细胞介素15(IL-15)是一种促炎性蛋白,在中性粒细胞、树突状细胞和巨噬细胞的活化中起作用,对于NK细胞和CD8+T细胞的发育和生存是必不可少的。IL-15通过异三聚体受体信号激活Jak/Stat途径。虽然IL-15mRNA在许多细胞类型中表达,但是该分泌蛋白在生物样品中很少检测得到。最近的研究表明,IL-15滞留在胞内,只有当与其唯一的受体IL-15Rα形成复合物才能分泌到胞外。可溶性IL-15/IL-15R复合物由小鼠树突状细胞、成纤维细胞和巨噬细胞产生,它的结合活性比单独的IL-15提高10-100倍。
