检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人IL-1β抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品和待测样本,经过孵育,样本中存在的IL-1β与固相抗体结合。洗涤去除未结合的物质后,加入生物素化的检测抗体孵育。洗涤去除未结合的生物素化的抗体,加入辣根过氧化物酶标记的链霉亲和素 (Streptavidin-HRP)。洗涤后,加入信号增强剂孵育,洗涤去除未结合的物质后,再次加入Streptavidin-HRP。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中IL-1β的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

IL1B 分子靶点综述

白细胞介素1(IL-1)是17kDa的胞外多肽,分为IL-1α和IL-1β两种蛋白。IL-1β是活化的巨噬细胞分泌的蛋白前体,通过caspase1(CASP1/ICE)水解产生的活化形式。该细胞因子是炎症反应的重要介导者,参与许多细胞活性,包括细胞增殖、分化和凋亡。IL-1是一类重要的蛋白家族,它们由生物活化的单核细胞所衍生,参与炎症反应和免疫应答。IL-1β表达升高会产生各种不同的自身炎症综合征,最显著的是单基因条件称为CAPS,是由于触发IL-1β加工的炎性体受体NLRP3发生突变而引起的。本试剂盒用来检测人IL-1β的成熟形式和未裂解的前体形式。
