检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人CXCL12抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的CXCL12与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中CXCL12的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

CXCL12 分子靶点综述
趋化因子CXC配体12(CXCL12)也称为基质细胞衍生因子1(SDF-1),是一种可活化白细胞的趋化因子蛋白,通常由促炎性刺激剂诱导产生,如LPS、TNF或IL-1。CXCL12对淋巴细胞具有强烈的趋化作用。在胚胎发生期,它可促使造血细胞从胎肝向骨髓迁移和大血管的形成。在成年人中,CXCL12在血管生成中具有重要作用,它通过CXCR4依赖的机制招募骨髓中的内皮祖细胞(EPCs)。CXCL12对肿瘤转移也有作用,表达CXCR4受体的癌细胞可被吸引至靶组织。然而在乳腺癌中,CXCL12表达升高则提示远端转移的风险降低。

Enables CXCR chemokine receptor binding activity; chemokine activity; and integrin binding activity. Involved in several processes, including integrin activation; negative regulation of apoptotic process; and regulation of leukocyte migration. Acts upstream of or within positive regulation of T cell migration and positive regulation of cell adhesion. Located in extracellular exosome. Part of collagen-containing extracellular matrix. Implicated in breast carcinoma; diabetic angiopathy; human immunodeficiency virus infectious disease; and type 1 diabetes mellitus. Biomarker of diabetic retinopathy; insulinoma; periodontitis; and type 2 diabetes mellitus.
