核因子-KB(Nuclear factor-kappa B, NF-KB)蛋白家族是一种多效的转录因子,可以与多种启动子部位的KB位点发生特异性结合,从而促使其转录表达。其受氧化应激、细菌脂多糖、细胞因子等多种刺激而活化后,调控前炎症性细胞因子、细胞表面受体、转录因子、粘附分子等生成。

NNFkB主要存在于细胞质中,并与抑制性的IkB蛋白形成复合物,当IkB磷酸化之后,NFkB被释放,并转位到细胞核。NFkB在凋亡和细胞存活上有着矛盾的功能,NFkB通过抑制p53依赖的凋亡和上调Bcl-2家族的抗凋亡成员和caspase抑制剂(如XIAP和FLIP),介导了许多信号的存活反应。相反,NFkB也被DNA损伤中的凋亡刺激所激活,介导了促凋亡基因(如TRAILR2/DR5、Fas和Fas配体)的上调。
