类风湿性关节炎(RA)是一种自身免疫性疾病。早期的类风湿性关节炎一般首先影响到小关节,随着病情的发展,症状蔓延到手腕、膝盖、脚踝、肘部、臀部和肩膀,最终导致骨侵蚀和关节畸形。RA微环境往往伴随着高度炎症、内源性气体水平失衡和治疗不力的恶性循环。为了打破这个循环,重庆大学、成都中医药大学、重庆医科大学附属第一医院的Wenbo Geng、Jie Zhao 、Bailong Tao等团队开发了一种双气体介导的可注射水凝胶,用于调节RA的免疫微环境,同时释放治疗药物。该研究成果在Bioactive Materials(IF=18.9)在线发表了题目为“Regulation of rheumatoid arthritis microenvironment via a self-healing injectable hydrogel for improved inflammation elimination and bone repair”的研究论文,此论文使用了联科生物Mouse IL-6 ELISA Kit(货号:EK206)、Mouse TNF-α ELISA Kit(货号:EK282)和Mouse IL-10 ELISA Kit(货号:EK210)3种检测试剂盒。
本文使用的水凝胶(DNRS凝胶)可以通过消耗过量的一氧化氮(NO)和释放治疗性硫化氢(H2S)按需分解,导致内源性气体恢复、炎症缓解和巨噬细胞极化为M2型。此外,水凝胶还可以抑制破骨细胞生成,增强成骨能力。在胶原诱导的关节炎大鼠模型中,关节内注射含有甲氨蝶呤的水凝胶(MTX/DNRS凝胶)显著减轻了炎症和临床症状,并促进了骨侵蚀的修复。体内结果表明,MTX/DNRS凝胶恢复了微环境,提高了MTX的治疗效果。该研究为开发用于RA治疗的多功能智能递送平台提供了新的思路。
