2024年4月2日,武汉大学张先正教授团队在Advanced Materials ( IF 29.4) 上发表了题为:Bacterial Biohybrids for Invasion of Tumor Cells Promote Antigen Cross-Presentation Through Gap Junction 研究论文。此论文使用了联科生物2种ELISA检测试剂盒对小鼠TNF-α和IFN-γ蛋白进行了定量检测:Mouse TNF-α ELISA Kit(货号:EK282)、MouseIFN-γELISA Kit(货号:EK280)。
由于肿瘤携带的细菌与宿主细胞在细胞组成和代谢行为上的固有差异,这些细菌可以有选择地影响肿瘤的免疫环境。然而,内源性细胞内细菌如何影响肿瘤细胞与抗原呈递细胞(APCs)之间的抗原呈递过程的机制尚不完全清楚。
本文研究了减毒沙门氏菌VNP20009(VNP)侵入肿瘤细胞的行为,并尝试通过修改VNP表面上的正电聚合物来调节这种行为。
研究发现:
①非毒性的壳聚糖寡糖(COS)修饰的VNP (VNP@COS)增强了VNP感染肿瘤细胞的能力,从而增强了肿瘤细胞与APCs之间的缝隙连接的形成。
②基于此基础,设计了一种细菌生物混合物,通过细胞内细菌诱导的缝隙连接促进原位抗原交叉呈递。
③这种细菌生物混合物还通过与VNP@COS结合的Mdivi-1增强了肿瘤细胞表面主要组织相容性复合体I类分子的表达。
④这种战略性整合有助于增强肿瘤抗原的免疫原性暴露,同时保留了T细胞的细胞毒性作用。提出了一种可以精确控制微生物在肿瘤内的功能和局部效果的策略。
