Notch通路是最基本的信号传导系统,使相邻的细胞相互调节、发挥其合适的发育作用。Notch蛋白通过细胞与细胞接触来介导重要的细胞功能。Notch信号传导包括调节胚胎和成年期多种细胞分化进程的基因调控机制。Notch受体是单次跨膜蛋白,包括功能性胞外区(NECD)、跨膜区(TM)和胞内区。在接收信号的细胞中,γ-分泌酶(也参与阿尔茨海默病) 可以在Notch 跨膜区的S3 位点水解,产生Notch 细胞内结构域(notch intracellular domain, NICD)。NICD转移入核与CSL (CBF1/Su(H)/Lag-1)形成复合物,从而激活Myc、p21和HES家族。
