ACE 分子靶点综述
血管紧张素Ⅰ转换酶(ACE),又称二肽基肽酶A,是一种锌金属肽酶,对血压控制和水盐代谢具有重要意义。ACE通过释放末端His-Leu二肽将血管紧张素I转化为血管紧张素II,从而增加血管紧张素的缩血管活性。它也能使缓激肽失活,后者是一种有效的血管扩张剂。ACE和ACE-2是两种细胞表面蛋白酶,是肾素-血管紧张素系统(RAS)的重要调节因子,在维持哺乳动物血压稳态和水盐平衡中起着关键作用。ACE以2种形态存在于人体中,一种表达于体细胞,称为sACE(somatic ACE),另一种表达于生殖细胞,称为gACE(germinal ACE)。2种形态的ACE主要不同点在于gACE只含有一个C端活性功能区,而sACE含有C端和N端2个活性功能区。因此,2种ACE的催化活性不同,在最重要的RAS升压调节系统中,sACE有更加关键的作用。ACE-2由一个蛋白酶结构域组成,是心肺功能的重要调节因子。它也是引起严重急性呼吸综合征的SARS病毒以及新冠病毒刺突蛋白的细胞受体。
